QUANTITATIVE KARYOTYPE OF DIPLODON CHILENSIS (GRAY 1828) (BIVALVIA:HYRIIDAE) Gayana
Jara-Seguel,Pedro; Peredo,Santiago; Palma-Rojas,Claudio; Parada,Esperanza; Lara,Gladys.
The karyotype of a freshwater population of Diplodon chilensis located in La Poza area of the Villarrica Lake (39º18'S; 72º05'W), Southern Chile, was studied. The chromosomes were obtained by squash of cleaving embryos, previously treated with colchicine and fixed in ethanol-acetic acid 3:1 at 4ºC and stained using the Feulgen reaction. Diplodon chilensis showed a diploid karyotype of 2n = 34, with metacentric and submetacentric chromosomes. This work is the first approach to cytogenetic characterization of Chilean populations of this species.

A new karyotype of Calomys (Rodentia, Sigmodontinae) Iheringia, Sér. Zool.
Lima,J. Fernando de S.; Kasahara,Sanae.
The genus Calomys Waterhouse, 1837 is widely distributed within South America, being found in Venezuela, Colombia, Peru, Bolivia, Brazil, Paraguay, Uruguay and Argentina. Specimens of Calomys were collected in Formoso do Araguaia, Tocantins, Brazil. For chromosome characterization standard staining techniques and as G-banding and nucleolar organizer region were used. The karyotype was 2n=46 and AN=66. The X chromosome is a medium metacentric and the Y chromosome a small acrocentric chromosome. Chromosome homologies with other species were observed. Probably, karyotype differences were basically due to Robertsonian rearrangements.

Evidence of differential chromosome loss in aneuploid karyotypes of the Pacific oyster, Crassostrea gigas ArchiMer
Leitao, Alexandra; Boudry, Pierre; Thiriot Quievreux, Catherine.
The G-banding technique was performed on aneuploid karyotypes from gill tissue of the Pacific oyster, Crassostrea gigas, to assess whether chromosome losses could be explained by differential chromosomal susceptibility and to clarify the negative correlation between aneuploidy and growth rate previously reported in different populations of this oyster. The study of 95 G-banded aneuploid karyotypes showed that only 4 of the 10 chromosome pairs (viz. 1, 5, 9, and 10) of C. gigas were affected by the loss of one homologous chromosome. Pairs 1, 9, and 10, which were lost in 56, 33, and 44% of cases, respectively, may be considered to be differentially affected. Hypotheses on this differential chromosomal susceptibility are discussed.

Acute promyelocytic leukemia: the study of t(15;17) translocation by fluorescent in situ hybridization, reverse transcriptase-polymerase chain reaction and cytogenetic techniques BJMBR
Chauffaille,M.L.L.F.; Figueiredo,M.S.; Beltrani,R.; Antunes,S.V.; Yamamoto,M.; Kerbauy,J..
Acute promyelocytic leukemia (AML M3) is a well-defined subtype of leukemia with specific and peculiar characteristics. Immediate identification of t(15;17) or the PML/RARA gene rearrangement is fundamental for treatment. The objective of the present study was to compare fluorescent in situ hybridization (FISH), reverse transcriptase-polymerase chain reaction (RT-PCR) and karyotyping in 18 samples (12 at diagnosis and 6 after treatment) from 13 AML M3 patients. Bone marrow samples were submitted to karyotype G-banding, FISH and RT-PCR. At diagnosis, cytogenetics was successful in 10 of 12 samples, 8 with t(15;17) and 2 without. FISH was positive in 11/12 cases (one had no cells for analysis) and positivity varied from 25 to 93% (mean: 56%). RT-PCR was done...

Differentiation of mouse embryonic stem cells and their hybrids during embryoid body formation Genet. Mol. Biol.
Mittmann,Josane; Kerkis,Irina; Kawashima,Cinthia; Sukoyan,Marina; Santos,Enrico; Kerkis,Alexandre.
We studied the karyotypes of three hybrid clones of mouse embryonic stem cells and murine splenocytes (two having near diploid and one having near tetraploid chromosome numbers) and the characteristics of their differentiation during the formation of embryoid bodies. The X chromosome originating from embryonic stem cells may be lost in hybrids with a near diploid chromosome number and reprogramming of the "somatic" X may occur. The morphological data we obtained using light and electron microscopy revealed a correlation between the karyotype constitution of hybrid cells and their differentiation during the formation of embryoid bodies. At the beginning of development, the embryoid bodies derived from hybrid cells already showed an advanced degree of...

Trans-Atlantic distribution of a mangrove oyster species revealed by 16S mtDNA and karyological analyses ArchiMer
Lapegue, Sylvie; Boutet, Isabelle; Leitao, Alexandra; Heurtebise, Serge; Garcia, P; Thiriot Quievreux, C; Boudry, Pierre.
Three species of man.-rove oysters, Crassostrea rhizophorae, C. brasiliana, and C. gasar, have been described along the Atlantic shores of South America and Africa. Because the distribution of these molluscs is of great biological and commercial interest, their taxonomy and distribution deserve further clarification. Therefore, 15 populations were sampled from both continents. Their 16S mitochondrial polymorphism was studied by sequencing and PCR-RFLP analysis. Two haplotypes were identified. Haplotype a was the only one observed in Africa, but it was also observed in South America together with haplotype b. Because C. gasar is the only mangrove oyster identified on the west coast of Africa, haplotype a was attributed to this species, which has thus been...

Cytogenetic data of Partamona peckolti (Hymenoptera, Apidae, Meliponini) by C banding and fluorochrome staining with DA/CMA3 and DA/DAPI Genet. Mol. Biol.
Brito,Rute Magalhães; Caixeiro,Ana Paula de Almeida; Pompolo,Silvia das Graças; Azevedo,Giselle Garcia.
The stingless bees of the Partamona genus have been studied taxonomically, ecologically and behaviourally, but cytogenetic studies are still rare. The objective of this study was to obtain cytogenetic data to contribute to Partamona peckolti species characterization. Heterochromatin was localized in all chromosome pericentromeric regions but some blocks could be visualized on some large chromosomes arms. A large heterozygous DA-CMA3-positive band was observed on one large chromosome arm, but was completely absent when C banding was applied before fluorochrome staining, with only one small positive band being visualized. Sequential DA-CMA3-NOR staining of interphase nuclei provided coincident positive responses. This suggests that DA-CMA3-positive bands of...

Karyotypes of two cytotypes of Paspalum quadrifarium Lam. (Poaceae): an alternative technique for small chromosomes in plants Genet. Mol. Biol.
Speranza,Pablo; Vaio,Magdalena; Mazzella,Cristina.
Paspalum quadrifarium Lam. is a bunchgrass native to Uruguay, Argentina, and southern Brazil. Diploid, triploid, tetraploid and hexaploid cytotypes have been reported for this species of the Quadrifaria group of Paspalum. In this group, a high degree of cytogenetic homology between the genomes of several diploid species has been reported, based on meiotic pairing in interspecific hybrids; multivalent associations would thus be expected in polyploid hybrids. Karyotype analysis could provide useful information about the genomic architecture of polyploid plants; however, the fully condensed mitotic chromosomes of Paspalum do not provide enough morphological features for such an analysis. In this paper, we used mitotic prometaphase chromosomes treated with 70%...

Multiple interstitial ribosomal sites (NORs) in the Brazilian squirrel Sciurus aestuans ingrami (Rodentia, Sciuridae) with 2n = 40: an overview of Sciurus cytogenetics Genet. Mol. Biol.
Fagundes,Valéria; Christoff,Alexandre Uarth; Amaro-Ghilard,Renata Cecília; Scheibler,Daniel R.; Yonenaga-Yassuda,Yatiyo.
This is the first time the karyotype of the Brazilian squirrel Sciurusaestuans ingrami, with 2n = 40, is described. The karyotype of this species comprises 18 pairs of biarmed and one minute pair of acrocentric autosomes, a medium-sized submetacentric X and a medium-sized acrocentric Y. Four pairs have an interstitial secondary constriction, co-located with nucleolar organizer regions (NORs), identified by silver-staining technique and fluorescent in situ hybridization (FISH) with ribosomal 18S/28S probes. The occurrence of multiple interstitial NORs is rare in rodents, and this is one of the few examples, identified by a molecular cytogenetics approach.

A new species of Calomys Waterhouse (Rodentia, Sigmodontinae)from the Cerrado of Central Brazil Zoologia
Bonvicino,Cibele R.; Lima,José F. S.; Almeida,Francisca C..
A new Brazilian Calomys Waterhouse, 1837 species is described based on morphologic and karyologic data. This species is endemic to the Cerrado of Central Brazil and allopatric with all other species of the genus Calomys. Its chromosome complement (2n = 46, AN = 66) is different from those described in other Calomys species. Morphometric analysis significantly distinguished this new species from other Calomys of the Brazilian fauna like C. callosus (Renger, 1830), C. expulsus (Lund, 1841) and C. tener (Winge, 1887) and placed it among the large-sized Calomys.

Karyotype of cryopreserved bone marrow cells BJMBR
Chauffaille,M.L.L.F.; Pinheiro,R.F.; Stefano,J.T.; Kerbauy,J..
The analysis of chromosomal abnormalities is important for the study of hematological neoplastic disorders since it facilitates classification of the disease. The ability to perform chromosome analysis of cryopreserved malignant marrow or peripheral blast cells is important for retrospective studies. In the present study, we compared the karyotype of fresh bone marrow cells (20 metaphases) to that of cells stored with a simplified cryopreservation method, evaluated the effect of the use of granulocyte-macrophage colony-stimulating factor (GM-CSF) as an in vitro mitotic index stimulator, and compared the cell viability and chromosome morphology of fresh and cryopreserved cells whenever possible (sufficient metaphases for analysis). Twenty-five bone marrow...

Karyotypic description of four species of Trigona (Jurine, 1807) (Hymenoptera, Apidae, Meliponini) from the State of Mato Grosso, Brazil Genet. Mol. Biol.
Costa,Kleber França; Brito,Rute Magalhães; Miyazawa,Carlos Suetoshi.
The genus Trigona contains at least 31 species, but there have been few cytogenetic studies of this group. In this work, four species of Trigona (T. branneri, T. chanchamayoensis, T. hyalinata, and T. recursa) from the municipality of Cuiabá, in the State of Mato Grosso, Brazil, were studied. In all of the species, the females had 2n = 34 chromosomes and the males had n = 17. The C-banding patterns showed that the karyotypes of these species consisted mainly of acrocentric and pseudoacrocentric chromosomes. These cytogenetic findings should useful in future phylogenetic studies of this group.

Cytogenetic analysis of five species of the subfamily Corydoradinae (Teleostei: Siluriformes: Callichthyidae) Genet. Mol. Biol.
Shimabukuro-Dias,Cristiane Kioko; Oliveira,Claudio; Foresti,Fausto.
In the present study, five callichthyid species belonging to the subfamily Corydoradinae were karyotyped: three species of Aspidoras and two of Corydoras. The three species of Aspidoras had the same diploid number, 2n = 46 chromosomes, similar karyotypic formulae, with most chromosomes metacentric or submetacentric, single interstitial Ag-NORs and C-band positive segments mainly found in the centromeric position. The comparative analysis of cytogenetic data available for the genus Aspidoras and other species of Corydoradinae suggest that several events of centric fusion occurred in the origin of the species of Aspidoras. The two analyzed species of Corydoras showed high diploid numbers, 2n = 74 in C. sodalis and 2n = 90 in C. britskii. While C. sodalis...

Chromosomal similarity between the Scaly-headed parrot (Pionus maximiliani), the Short-tailed parrot (Graydidascalus brachyurus) and the Yellow-faced parrot (Salvatoria xanthops) (Psittaciformes: Aves): a cytotaxonomic analysis Genet. Mol. Biol.
Caparroz,Renato; Duarte,José Maurício Barbanti.
Behavior, morphology, allozyme studies and DNA hybridization and sequencing data all suggest the independent evolution of the Old and New World parrots and support tribe status for the American species, although the phylogenetic relationships within this tribe are still poorly understood. A previous study has shown that the Yellow-faced parrot (Amazona xanthops Spix 1824) exhibits large karyotypic differences compared to the other Amazona species and suggested that this species should be renamed Salvatoria xanthops, although the relationships between S. xanthops and the other New World parrots remain unclear. Cytogenetics of three Brazilian species of Eleutherodactylus (Anura, Leptodactylidae) with 22 chromosomes and re-analysis of multiple translocations in E. binotatus Genet. Mol. Biol.
Siqueira-Jr.,Sérgio; Ananias,Fernando; Recco-Pimentel,Shirlei M..
In this paper, we provide a cytogenetic analysis of Eleutherodactylus guentheri, E. parvus and E. binotatus. All of the species had a diploid chromosomal number of 2n = 22. The karyotypes of E. guentheri and E. parvus were very similar and differed only slightly in the morphology of pair 2. These two species also had an NOR-bearing secondary constriction on the long arms of pair 6. The karyotype of E. binotatus differed from those of E. guentheri and E. parvus in the morphology and size of the chromosomes, in the number of chromosomal arms, in the NOR location (detected on the short arms of pair 1), and in the pattern of heterochromatin. These results reinforce the differences between E. guentheri and E. binotatus and support the existence of two species...
